An EMSA is an electrophoretic mobility shift assay, also referred to as a gel shift assay, band shift assay, or gel retardation assay. EMSA is used to study protein-DNA interactions or protein-RNA interactions. Standard methods for studying transcription factors and gene regulation include EMSA, western blots and reporter gene assays, which are time-consuming, do not allow for high-throughput and provide only semi-quantitative results. Alpha Innotech's Chemiluminescent FAST Transcription Factor Microplate Assays employ a proprietary assay technology and chemiluminescent detection that offers superior sensitivity, broad dynamic range and excellent reproducibility compared with traditional methods. In addition, our FAST assays do not require the use of radioactivity and results can be obtained in under four hours, unlike time-consuming commercially available EMSA.
Standard methods for studying transcription factor and gene regulation include gelshifts, western blots and reporter gene assays, which are time-consuming, do not allow for high-throughput and provide only semi-quantitative results. Alpha Innotech's Chemiluminescent FAST Transcription Factor Microplate Assays employ a proprietary assay technology and chemiluminescent detection that offers superior sensitivity, broad dynamic range and excellent reproducibility compared with traditional methods or with commercially available transcription factor ELISAs. The assays do not require the use of radioactivity, and results can be obtained in under four hours. The convenient stripwell format enables testing of from one to 96 samples in each plate. Chemiluminescence is used to measures the binding of activated the transcription factor to their cognate DNA binding sequences.

A protein array is widely becoming the platform chosen by researchers for quantitative determination of serum levels. A protein array can be spotted with target peptides, antibodies or complex mixtures utilizing protocols reminiscent of western blot and ELISA. Antibody microarrays exist in two general types: using matched antibody pairs for sandwich-type and relying on single printed antibodies and a direct sample labeling methodology. Cell and molecular biologists interested in measuring protein array levels can utilize standard slide formats or can scale up to 96 well plates to screen hundreds or thousands of samples. Some key requirements are multiplex detection utilizing multiple antibody tags, flexibility in adopting new dyes for specific antibody epitopes and high resolution imaging of high density spots with a protein array.
A gel shift assay, also referred to as an EMSA is an electrophoretic mobility assay. Gel shift is used to study protein-DNA interactions or protein-RNA interactions. Standard methods for studying transcription factors and gene regulation include gel shift, western blots and reporter gene assays, which are time-consuming, do not allow for high-throughput and provide only semi-quantitative results. Multicolor westerns are an ideal way to simultaneously detect multiple proteins closely related in size. Likewise, multicolor westerns allow evaluation of the phosphorylated and/or unphosphorylated state of proteins. Using antibodies with fluorophore conjugates, such as Quantum dots (Qdots), both sets of antibodies can be detected. The Alpha Innotech FluorChem family of imaging platforms integrates multiple filters into the emission filter wheel included in each darkroom component of the system. These filters allow researchers to target specific fluorophores, like those commonly used in multicolor westerns. Each fluorophore can be detected individually in a TIF file, and image files can be overlayed to present the detection of more than one fluorophore per sample. The FluorChem family of images is an ideal system to detect multicolor westerns.
